Background

DPCP (diphencyprone) is een witte kristallijne substantie. Het is een obligaat contactallergeen: sensibilisatie treedt in principe op na het eerste huidcontact. Voor zover bekend bestaat er geen kruisreactie met andere allergenen in de natuurlijke of technische omgeving van de mens. DPCP lost niet op in water, maar wel in aceton, ethanol of cyclohexane. De stof ontleedt door UV straling. Afgeschermd van zonlicht is de oplossing stabiel bij kamertemperatuur. Het smeltpunt ligt op 119-120° C. DPCP is hoog-reactief, een voorwaarde voor een potent contact allergeen. Hierom is het niet een stof voor industriële toepassing, maar wel voor topicale immuno-therapie.

Ames test
The Ames test (bacterial plate incorporation assay) has been carried out, by use of various Salmonella thypimurium strains, in two different laboratories (Prof.dr.H.Marquardt, Institute of Pharmacology, University of Hamburg; Department of Experimental Toxicology, Schering AG, Berlin). The results obtained show that DPCP displays no mutagenicity in the Ames test, both in the absence and presence of mammalian microsomes (host-mediated assay performed with S-9 mix).
 

Teratogenicity in the mouse
A teratogenicity study was performed in mice by Priv.Doz.Dr.B.Zimmermann (Institute of Toxicology and Embryonic Pharmacoly, Free University, Berlin). This study has shown that the substance, when applied in a dosage far higher that the dosage given per kg in humans, has no teratogenic effect on murine embryos. Two series of ten pregnant NMRI mice were treated either with 1 mg DPCP/kg or 10 mg DPCP/kg, dissolved in 1,2-propanediol (0.2 ml/40 g mouse). At a dosage of 1 mg/kg there was no fetal damage, and the average birth weight of the fetuses was unchanged. At a dosage of 19 mg/kg the fetal birth weight was significantly reduced. The frequency of defects was 5,5%, being only slightly above the irrelevant level of 5%.

 
Hen's egg test
DPCP was investigated in the hen's egg test by Priv.doz.Dr.N.P.Lüpke (Institute of Pharmacology, University of Münster). The following dosages were tested: 0.1 mg/egg, 0.5 mg/egg, 2.0 mg/egg, 10.0 mg/egg. The substance did not show any teratogenic effect even at a dosage of 10 mg/egg, exceeding by far the dosage applied per kg in humans. The hatched chickens were examined for the following blood parameters: total serum protein, creatinine, glucose, uric acid, cholesterin, triglycerides, total bilirubin, GOT, GPT, LDH, alkaline phosphatase, gamma-GT, sodium, potassium, calcium chloride, anorganic phosphate. All of these parameters remained unchanged when compared to the control group.

Uses

DPCP is toepasbaar bij mensen omdat het niet aanwezig is in het natuurlijke of technische milieu van de mens, zodat ongewenste eczemateuze reacties vermeden worden, geen kruisreacties vertoont met andere stoffen die in het natuurlijke of technische milieu aanwezig zijn, uit toxicologisch oogpunt geschikt is voor langdurige lokale toepassing (zie onder).
DPCP wordt gebruikt voor het kunstmatig opwekken van een contactallergie bij mensen en is een veilig alternatief voor l-chloor-2,4-dinitrochlorobenzeen (DNCB) en squaric acid dibutyl ester (SADBE). Therapeutisch wordt DPCP toegepast als één van de behandelingsmodaliteiten van alopecia areata. DPCP is ook toegepast bij therapieresistente verruca vulgaris of plantaris.

 

Cross-Reactions
Voor zover bekend bestaat er geen kruisreactie met andere allergenen in de natuurlijke of technische omgeving van de mens.
